Frozen shoulder
What s your experience with frozen shoulder in menopausal women? @Anthony Castore

From my experience, frozen shoulder is a symptom of low estrogen. Just adjusting estradiol and progesterone levels should greatly improve it.
Pinealon and epithalon
I'm 50 years old and I sleep really well already. Usually get about 2 hours of REM and 2 hours of DEEP *min 20% of each. But I'm considering using Epithalon and/or Pinealon for the longevity and cognitive benefits. Would it make sense at all or wouldn't be needed?? Thanks!

The Brain Energy Reset: Ketones, NAD+, and Neuronal Resilience
Most people treat the brain like a furnace. When it runs cold, when the focus thins out and the afternoon turns to fog, the instinct is to shovel in more fuel. More coffee. More sugar. More of whatever promises a lift. The logic feels airtight. Low energy means add energy. But the brain is not really a furnace. It is closer to a house run by a thermostat, and most of the time the problem is not the size of the fire. It is the quality of the signal telling the system what to do. Hold onto that picture, because almost every mistake people make with brain energy comes from confusing the furnace with the thermostat. Start with the fuel, because that part is real. Your brain runs on two main substrates. The default is glucose, sugar pulled from the blood and burned for quick energy. Glucose is the loud, on-demand generator in the basement. It works, it is always running, and it is also a little dirty. Lean on it too hard for too long and the output gets noisy. The 3pm wall, the flat afternoons, the sense that you are revving the engine just to hold a steady line, a lot of that is a brain stuck running its loudest fuel with nothing quieter to switch to. There is a second line, and most people never bring it online. Ketones. The main one worth knowing is beta-hydroxybutyrate, which everyone shortens to BHB. Here is the part that gets misunderstood. BHB is a cleaner fuel, and that is true, but the cleaner burn is not the headline. The headline is that BHB barely behaves like fuel at all. It behaves like a message. When circulating BHB sits in roughly the half to two-and-a-half millimolar range, the kind of mild ketosis you reach through fasting, low-carb eating, or exogenous ketones, it does two things that have nothing to do with combustion. This is where the biology is genuinely well established, so I will say so plainly. First, BHB acts as an HDAC inhibitor. Unpack that. HDACs are little molecular hands that keep certain genes wound up tight and switched off. When BHB blocks them, those genes get to unspool and turn on. One of the genes that comes online is FOXO3, which you can think of as the foreman that calls up your antioxidant defense crew. So a molecule you assumed was just fuel walks into the nucleus and tells the cell to build its own protection. Second, BHB quiets the NLRP3 inflammasome. The inflammasome is the brain's smoke alarm, a sensor that, when it goes off, drives the kind of low, smoldering neuroinflammation that wears down neurons over time. BHB turns the sensitivity down. So the cleaner fuel arrives carrying two instructions at once. Protect yourself, and stop sounding the alarm. That is the furnace log that is also a hand on the thermostat.

Love the content! What do you think of the strategy of running 5 Amino 1MQ before taking precursors? This would clean out accumulated NNMT and then the system would be better suited to utilize the precursors more efficiently, I would assume. After an initial cycle of 4 weeks of 5 Amino would it make sense to use precursors in conjunction with 5 Amino or keep them separate and lay off of the 5 Amino? Also, should we feel something when using 5 Amino? I used it for 4 weeks at a lower dose of 500mcg but never felt anything from it in terms of additional energy. I'm very lean already, about 10% BF and 125lbs *5,7 height. I tried a higher dose of 1mg but again, no effect so I lowered it back again because I believe that more is not always better. Should I try again using higher doses or is it doing its job despite me not having apparent higher energy levels? I'm already pretty energetic and have no problems with generating energy for high intensity efforts nor have any energy drops in the afternoon. I feel a steady influx of energy all day long. I'll add MOTS C to my stack at low doses as well, starting at 200mcg. Main intention is to keep the machine optimized and longevity. Wouldn't mind having extra energy though :)!

@Anthony Castore @Anthony Castore thanks for the answer! You’re very kind with your time and I love your explanations! Would you say that it’s still worth it for me to use 5 Amino? I still have some but I’m on a break now. Use precursors on the day that I use 5 Amino? Take MOTS separately so I’m able to determine what gave the effect; got it! Do you advise to adjust the dosage of MOTS based on specific symptoms like higher perceived energy? I started at 200mcg and I’m using it every other day. Thanks again!
SGLT2 and Trehalose
Hello everyone! There’s been much talk about SGLT2 inhibitors and its benefits in terms of longevity and overall optimization. I’m really interested in trying it. Has anyone tried it and what are your experiences? My concern in my situation is that I don’t eat a lot of carbs and my fasting insulin is already low, below 3. Wouldn’t that give me the risk of having eu glycemic keto acidosis? Any insights? About Trehalose, I read Anthony recommending taking it fasted for autophagy. Great. However, taking it fasted wouldn’t it break the fast since it’s an actual sugar? And for the autophagy activating benefits what dose should be taken at once?

@Anthony Castore hello Anthony! Wow, thank you só much for the explanation. I’m using spermedine at low 1mg doses but I was favoring using it at night after learning about its circadian rhythm influencie as well. And considering that we enter a clean up remodeling and recovery phase at night I thought that it may bring interesting benefits as well. Now I’m considering that doing a split dose may be an interesting option or would that be overshoot? I train fasted at 8am. Since training js already an autophagy inducing state, maybe it would make more sense to take it fasted after training? I usually eat about 60 min after training so that’s when I’m entering my Mtor activation phase. As far as Trehalose, I’m considering taking it after eating. From my understanding it would still give me benefits in terms of microbiome and cellular signaling. Do you believe it’s a good strategy taking Trehalose with coffee after lunch? Thanks again! Allulose and trehalose
What is Anthony’s opinion on allulose? I know he recommends trehalose, which I’ve also been using lately. However, I’ve been reading a lot of positive things about allulose and its effects on metabolic pathways in the body. What is the difference in their effects in this context? I’m not referring to carbohydrate content, calories, or sweetness, but rather their impact on fat metabolism, glycogen storage, inflammatory processes, and autophagy etc

Love this post and I have much interest in trying Trehalose. You recommend taking it fasted to enhance autophagy. However; since it’s a sugar wouldn’t it break the fast?
